Research Interests:

The research field of our laboratory is based on gastrointestinal physiology. The current research project mainly discusses gut microbiome and HBV transinfection and the gut-liver axis communication. Integration of metagenomics, GI physiology and liver innate immune profiling has provided important insights into the functions of HBV-gut microbiome to examine the hypothesis that the close linkage between gut microbiota and the persistence or clearance of HBV will be explored mechanistic insights into the specific phylum targeting the gut microbiota. Our emerging understanding of interplay between gut microbial dynamics and host innate immunity responses will further the development of effective interventions.

Our laboratory uses the in vivo and in vitro experiments to study the gut-liver axis related research through the animal model and cell molecular level of the disease. The laboratory has established HBV- HDI mice, Germ free mice, CRISPR-Cas9 TLR4 mice, TLR4 mutant mice, intestinal obstruction and other mouse models, and with a variety of molecular biology experimental techniques to further explore the role of cellular molecules. Experimental techniques include:

(1)  Intestinal, liver function and phenotype assay: ALT, AST, HBsAg, HBeAg, Anti-HBs, Anti-Hbe, Anti-Hbc, HBV DNA assay, bacterial translocation, H&E staining, isolation of total liver parenchymal and nonparenchymal cells and flow cytometry analysis.

(2)  Biochemical technology: immunofluorescent staining, surface and intracellular staining for flow cytometric analysis and sorting, fluorescent in situ hybridization, ELISPOT, Adoptive transfer, hydrodynamic injection.

(3)  Molecular biotechnology: RNA and DNA extraction, RT-PCR, siRNA knockdown, site directed mutagenesis, plasmid transfection, bacterial 16S rDNA sequencing, Temporal temperature gradient gel electrophoresis (TTGE), Southern Bolt, western blotting, Northern blotting. Gene expression profiles using NanoString or RNAseq.
